Either Mrs Pacman or a game called Lady in wading for the Atari - a gender reversed game where you play a female knight out to save her prince. Both were released in 1982 so take your pick.
Personally I’ve got to go with Mrs Pacman - simply because she’s still around.

‘Rougue’ (1980) doesn’t have an explicitly female protagonist, the adventurer’s gender is chosen by the player. Later rouguelike games (including one of my favourites, Nethack) have women only classes. Not sure if that counts

Sadly i’m not going to allow Rougue because it doesn’t have a specific female character.

The correct answer is Ms Pacman.
She was originally called Miss Pacman but when they introduced Pacman Jr the makers thought they should make an honest woman out of the lady Pacman.
Odd that she had the same last name as her future husband. Hope they weren’t related or anything. That’d be weird.
